Chord theory helps massively too, so work on dominant 7, major 7 and minor 7th chord arpeggios - they'll give you the root, third, fifth and seventh (if you choose to use it) of ANY chord in 95% of western popular music.

Have you checked all of the screws on the machine head? Check that the big screw through the centre of the tuner hasn’t worked loose, and also the 4 screws that hold the backplate to the headstock would be my next step- also, is there any ‘wobble’ in the tuning key itself when the string is tuned?

Tapes are MUCH wider in diameter than rounds, I’d start by checking the size of the groove for that string.
